Description:
(1) Epigenetic changes, such as DNA methylation and histone modification, cause abnormal NY-ESO-1 expression in the cytoplasm. (2) NY-ESO-1 is tagged with K48-linked polyubiquitin, directing it for proteasomal degradation. (3) The proteasome breaks it into peptides, which are transported into the ER by TAP. (4) These peptides bind to MHC class I molecules, moving to the surface for CD8+ T cell recognition.
