Anti-TNF Therapy in a Mouse Model of Rheumatoid Arthritis
Description
This template compares two soluble TACE constructs in a collagen-induced mouse model of rheumatoid arthritis: naked shTACE, which provides only moderate TNF-alpha regulation and leads to synovial inflammation, bone resorption, and joint destruction, and shTACE fused with the 8D-16R peptide, which blocks TACE cleavage of TNFR and significantly reduces inflammation and joint damage. Each pathway is traced at the joint cell membrane through synovial cells and monocytes down to MMP secretion and osteoclastogenesis.
