Description
PBMCs are treated with viral peptides, stimulated for 3 hours at 37°C. They are labeled with IFN-γ/CD45 antibody conjugates for 5 minutes on ice, then incubated for 45 minutes at 37°C to encourage cytokine secretion. A secondary monoclonal antibody against IFN-γ, along with PE, labels the capture matrix. Cells are stained with additional surface markers, incubated with anti-PE microbeads, and purified through two rounds of sorting on magnetic separator columns to enhance enrichment.
