Lysis – destroy the cell membrane/cell wall to denature proteins and release nucleic acids. This usually involves detergents.
Separation/Precipitation and Washing – selectively bind and precipitate/separate nucleic acids out of solution, separating them from cell debris, proteins, etc. This usually involves a reversible binding mechanism based on salt concentration
Elution – resuspend DNA in a buffer that forces it back into solution, allowing downstream use.
