Description:
This template shows a six-step ELISpot assay protocol in which antigen-secreting cells are incubated in antibody-coated wells, followed by capture of the secreted antigen, incubation with a biotin-conjugated detection antibody, and binding of alkaline phosphatase-conjugated streptavidin. A chromogenic substrate is then added, producing colored spots at the well base that correspond to individual antigen-producing cells.
