In short, they isolated splenocytes from the spleens of WT and FcmR KO mice. then they stimulated with a cocktail of substances towards B1 and B2 phenotypes. In the next step then transduced their hybridoma Mott cells (plasmacytoid cells that have prominent granules of IgM but secrete little IgM), with cDNA of missing FcmR.
With sorting and confocal microscopy, the proved rescued phenotype of WT Mott cells (fewer granules and more IgM).
