The multistage approach involves the use of a stimuli-responsive carrier, such as nanoparticles, to cause drug release in the tumor. The primary nanoparticle delivers the secondary nanoparticles containing the cytotoxic payloads to the tumor microenvironment via the enhanced permeability and retention effect. The primary nanoparticles break down into secondary nanoparticles and release drugs in the tumor microenvironment when exposed to acidic pH or particular enzymes.
