Systemic nanoparticle therapy suppresses breast tumors through RNA interference (RNAi) of long non-coding RNA DANCR. PEG and RGD peptide improve biocompatibility and tumor uptake of the ECO/siDANCR nanoparticles, respectively. After endosomal escape at low pH, the nanoparticles release siDANCR, which is used by RNA-Induced Silencing Complex (RISC) to degrade cellular DANCR, thus preventing tumorigenic chromatin changes, epithelial-mesenchymal transition (EMT), and phosphorylation of oncokinases.
