Description:

Depending on whether donor DNA for repairing the DSB is provided along with the ZFN, the DNA ends are ligated by NHEJ (absence of template DNA) or HDR (presence of template DNA). Conventionally, the process of NHEJ is accompanied by the loss or gain of nucleotides, resulting in disruption of the gene. In HDR, repair occurs in the presence of a DNA sequence homologous to the cleavage site, serving as a template for the synthesis-dependent strand annealing pathway.
