shRNA plasmid library with unique barcodes to genes of interest are cloned then transduced into tumor cell line using viral vectors. Following transduction, cells are injected and allowed to grow in mice. One group of mice receives treatment of interest, another is the untreated control. Following treatment, tumors are harvested and analyzed for barcodes using microarray; changes in the specific barcode levels between treated and untreated groups suggests higher or lower gene expression.
