Protocol to Culture Primary Human Basal Epithelial Cells in 2D and 3D System

An editable high resolution scientific image depicting Protocol to Culture Primary Human Basal Epithelial Cells in 2D and 3D System

Description

This template was adapted from the original submission. Edits were made to enhance scientific accuracy, optimal usability and/or to meet industry-leading design standards for science communication. Human airway biopsies are harvested by bronchoscopy. Biopsies are cut to small explants that are added on 3T3 mouse fibroblasts to allow human basal epithelial cells (HBECs) expansion. HBECs are further expanded on fibroblasts to increase their number (2D culture system). HBECs are then separated from fibroblasts by differential trypsinization. They are seeded in Matrigel, and immune cells added at one week of culture (if needed). Organoids are formed 20 days after culture (3D culture system).
