RNA Polymerase II: promoter clearance, promoter proximal pausing and productive elongation phase
Description
RNA Polymerase II binds to the DNA promoter region with help from the pre-initiation complex (PIC). After promoter clearance, RNA Pol II makes 20-60 nucleotides of mRNA, then pauses due to DSIF and NELF—this is called promoter-proximal pausing. To enter the elongation phase, p-TEFb must phosphorylate NELF, DSIF, and the C-terminal domain of RNA Pol II. This allows other factors like ELL-EAF and Elongin to suppress pausing, enabling RNA Pol II to continue with productive elongation.
