Description

In normal physiological conditions, MDM2 inhibits P53 activation, consequently preventing P21 activation. This inhibition enables the CyclinE/CDK2 complex to phosphorylate RB, into p-RB, freeing E2F1 to facilitate G1/S cell cycle progression. However, when DNA double-strand breaks (DSB) occur, P53 and P21 are activated. P21 then forms a complex with CyclinE/CDK2, inhibiting its ability to phosphorylate RB. Unphosphorylated RB in turn binds with E2F1, resulting in cell cycle arrest.
