p21-Mediated Inhibition of Cell Cycle Progression: p21 binds to and inhibits the CDK2-cyclin E complex, preventing ATP-driven phosphorylation of pRb. Hypophosphorylated pRb remains bound to E2F transcription factors, blocking the expression of genes required for DNA synthesis and cell cycle progression. Consequently, the cell enters arrest, halting division and DNA replication
