Role of PARP1 in DNA damage repair
Description
Poly(ADP-ribose) polymerase-1 (PARP-1) human enzyme, using NAD+ as the building block, catalyzes the creation of the branching polymer poly(ADP-ribose). PARP-1 play an important role in DNA damage repair. For instance, inactivation of PARP-1 by caspase-3 cleavage is one of the essential steps of apoptosis, whereas over-activation of PARP-1 results in ATP depletion and necrosis. In the event of modest damage, PARP can recruit other repair enzymes and DNA repair can occur.
