Description
SHAPE reagents react preferentially with the 2′-hydroxyl groups of conformationally flexible RNA nucleotides. Overview of the three most useful SHAPE reagents. 1M7 is the workhorse SHAPE reagent; its reactivity with RNA measures local nucleotide flexibility 1M6 and NMIA are selective for nucleobases that have one face available for stacking and that achieve a reaction-competent conformation on a slow timescale, respectively.
