Description

Chronic itch is linked to spinal circuit sensitization. We identified a microcircuit with Aβ-LTMRs, Ucn3⁺ excitatory, and NPY⁺ inhibitory interneurons mediating mechanical itch. Under chronic itch, Ucn3⁺ neurons became hyperexcitable, while NPY⁺ neurons were suppressed. qRT-PCR showed increased Nav1.6/Cav2.3 and decreased SK3 in Ucn3⁺ neurons, and reduced Nav1.6/BK in NPY⁺ neurons. Modeling revealed Nav1.6 changes as the main driver of circuit sensitization.
