Strategies to target IDH mutant tumors
Description
Mutant IDH1 (mIDH1) and mutant IDH2 (mIDH2) proteins catalyze the conversion of α-ketoglutarate(α-KG) to 2-hydroxyglutarate(2-HG). 2-HG is a competitive inhibitor of α-KG-dependent enzymes including Tet methylcytosine dioxygenases and jumonji domain-containing histone lysine demethylases, leading to DNA hypermethylation. In addition, mIDH1/2 may have other oncogenic effects driven by 2-HG.
