Description:
The Tango assay is a useful technique for studying GPCR activation and signaling. The receptor is fused to a transcription factor that will be translocated to the nucleus when the receptor is activated. Then, it will induce the expression of a reporter gene, such as eGFP (enhanced green fluorescence protein), allowing us to measure its signaling by flow cytometry. This technique is a great way of probing whether nanobodies or other molecules are capable of activating/deactivating GPCRs and how.
