Healthy cells gain unlimited replicative potential through hTERT activation or through gain of chr.8q24.3/TONSL amplification. Theses TONSL amplified cells have hyperactivated DNA repair pathway which lead to cellular imbalance. With gain of additional driver mutations theses TONSL amplified cells lead to formation of TONSL-dependent tumors. These TONSL dependent tumors can be targeted with CBL0137, a TONSL/FACT complex specific inhibitor.
