Description
An essay to screen treatment effects on T. cruzi amastigote intracellular growth in mammalian cells. For this assay parasites expressing the bacterial B-galactosidase (lacZ) (ATCC PRA-330) are treated and then inoculated on wells containing a monolayer of mammalian cells. The infection proceedes for 48 h and then cell lysis and addition of the galactosidase substrate CPRG (chlorophenol red‐β‐D‐galactopyranoside) allows for parasite load measurement by reading absorbance between 570 and 595 nm.
