In the IFN-γ pathway, after ligand binding IFN-γ receptor (IFNgR1 & IFNgR2) gets activated, and results close association of JAK1 and JAK2 proteins facilitate auto- and transphosphorylation and enzymatic activation. Activated JAK proteins phosphorylate the STAT1 binding site, activating STAT1 dimerization and translocation to the nucleus and it binds to gamma-activated site (GAS) elements and promotes gene transcription.
