Syngeneic tumor cell line models that are injected into immunocompetent hosts (commonly either subcutaneously or orthotopically) allow (a) testing of immunotherapy treatments, (b) in vivo screens to identify mediators of response and resistance to immune-oncology treatments, and (c) genetic manipulation to mimic resistance mechanisms found in patients. Abbreviations: NSG, NOD-Scid-gamma−/−; sgRNA, single-guide RNA; shRNA, short hairpin RNA.
